I have a t3500 motherboard and wanted to know if that CPU heat sink backplate comes off if so how

Most likely from the Torx screws that hold the CPU retention bracket on. If pulled, there would be no threaded holes for the retention bracket any longer.

yes it should come off, it might have double sided sticky pads on it.

The nub sticking through is not screwed into the back plate?
