But why would you open the distro fill port while the system is running? There's only a vacuum while the system (or at least the vacuum pump) is running after all. Fill the system properly, then activate the leakshield, and it should work.
@Shoggy specifically warned about possible issues arising from the process of arming the system, setting the appropriate dual pump pressures and the required vacuum balance, which more often than not could trigger fill level issues In both pump reservoirs.

Distro Plate reservoirs have a critical fill level requirement and air bubble contamination when the system is inadequately primed. I have no idea what to expect in such an instance with the EK Quantum Reflection PC-O11D XL D5 PWM. I would expect Aqua Computers to have tested pairing the leakproof system in various system possibilities.

According to @Shoggy i will need to adjust the fill levels manually, therefore I need to know the possibilities when:
  1. When combining a flat reservoir with the classic Cylindrical reservoir;
  2. Could the LeakShield be implemented when pairing the FLT Distro Plate with the D5 integrated airplex modularity system

You will have to replace the top of one reservoir with this one. The other reservoir will get LEAKSHIELD as its new top.
Both pumps together will have a to high pressure. You will have to slow them down.

The pressure of a pump increases quadratically with the speed.

H2=H1 * (n2/n1)²

H1 is the known pressure
n1 is the nominal speed
n2 is the set speed

For a D5 with H1 = 365 mbar and n1 = 4800 rpm, you can use this formula to calculate the max. pressure for each speed.
For example, if the pump is running at 3000 rpm, you calculate 365 mbar * (3000 rpm / 4800 rpm)² = 142.6 mbar.

So, with two pumps set to 3000 rpm, you will get a total pressure of about 280 mbar which is fine for LEAKSHIELD.

The filling feature of LEAKSHIELD could be a problem because the fill level in both reservoirs will end up differently for sure. It is likely that you will have to adjust this manually.
