HD Tune Pro measures performance across the whole capacity of the drive. This allows identifying weak spots in the architecture or media access. For the read tests the drives were filled up with random data. This ensures that actual data is read and the controller can not just return all zeros if it knows a block to be empty, without accessing any actual data.
